A SARS-CoV-2 spike-targeting bispecific T-cell engager strategy for controlling SARS-CoV-2 infection CommsBio covid COVID19 SARSCoV2 tcell spike strategy
By Dr. Chinta SidharthanJun 5 2023Reviewed by Lily Ramsey, LLM In a recent study published in the Communications Biology Journal, researchers described a novel strategy for controlling severe acute respiratory syndrome coronavirus 2 infections where a bispecific T-cell engager is used to target the SARS-CoV-2 spike protein.
Many of these antibody therapies target the angiotensin-converting enzyme-2 receptor, the primary receptor for SARS-CoV-2 entry. This fusion protein comprises an extracellular ACE-2 domain for blocking viral entry and an anti-CD3ε single-chain variable fragment to eliminate the virus-producing cells by activating T cells.
Furthermore, a pseudotyped SARS-CoV-2 production system was used to test the efficacy of S-BiTE in preventing viral release. The cytotoxicity of S-BiTE was also tested in vivo. Results The results reported that S-BiTE competed with membrane receptors and blocked the entry of free SARS-CoV-2 into permissive cells while activating strong T-cell mediated cytotoxicity to eliminate virus-infected cells.
