In the NATION-1907 trial, treatment of patients with resectable esophageal squamous cell carcinoma with the anti-PDL1 agent adebrelimab was safe and showed preliminary overall survival efficacy
. Although our bulk TCR-seq supports the first possibility for the source of tumor-reactive T cells induced by PD-L1 blockade, further work is required to identify the functional status–clonotype relationship of T cells using single-cell RNA-seq and TCR-seq.
These data are complementary to standard-of-care treatments in neoadjuvant settings. The translational study identified patients with favorable pathological responses to immunotherapy, supporting a promising alternative regimen to avoid overtreatment by chemotherapy and chemoradiotherapy with or without immunotherapy.
Neoadjuvant adebrelimab was discontinued if one of the following occurred: informed consent was withdrawn because of a patient’s personal decision, or there was unacceptable toxicity, disease progression according to RECIST 1.1 criteria, or a patient had a life-threatening disease or condition preventing further treatment. Safety and efficacy data were reviewed by an independent data-monitoring committee. There was no bias toward age, gender or race in this clinical trial.

Chemical generation of checkpoint inhibitory T cell engagers for the treatment of cancer - Nature ChemistryThree-protein conjugates, which have so far been produced using protein-engineering strategies, can now be generated using a chemical approach that enables the addition of small-molecule functionality. Checkpoint inhibitory T cell engagers (CiTEs) were assembled and shown to have enhanced in vitro potency compared to a traditional T cell engager.

Correlated insulator collapse due to quantum avalanche via in-gap ladder states - Nature CommunicationsThe microscopic mechanism of the electric-field-driven insulator-metal transition in strongly correlated systems has been debated. Here the authors present a general theory based on a quantum avalanche mediated by the formation of in-gap ladder states from multiple-phonon emission.
