More than 3,000 quails, 1,000 chukars and 100 chickens died in the blaze in Greenup County, Kentucky.
A number of dogs are also reported to have been killed in the fire.
One of the barns involved in the fire, the Bonso Barn, was more than a century old, according to the family that owns it.
