A case series showed that chimeric antigen receptor T-cell therapy targeting CD19 halted the progression of fibrotic organ manifestations in six patients with diffuse cutaneous systemic sclerosis.

CD19-targeting chimeric antigen receptor T-cell therapy shows potential to intercept fibrotic organ manifestations and improve disease measures in patients with diffuse cutaneous systemic sclerosis who had disease progression despite multiple previous treatments.

The secondary outcomes included changes in the modified Rodnan skin score , imaging and laboratory assessments, patient-reported outcomes, and the modified American College of Rheumatology Composite Response Index in Systemic Sclerosis , assessed at baseline and 3, 6, 9, and 12 months after treatment.No progression of organ manifestations or new lung, cardiac, or renal events occurred within the median follow-up period of 487 days.

Skin involvement improved in all the patients after CAR T-cell treatment, with a median mRSS decrease of 8 points within 100 days; the improvements were maintained throughout the 1-year follow-up period.IN PRACTICE:
